3:00-4:00 pm (Tokyo)
5:00-6:00 pm (Melbourne)

Join us from 2:00-3:00 pm (Singapore Time) on 22 March 2022 for the PacBio Asia Pacifc webinar featuring Prof. Yun Liu of Fudan University.to find out how HiFi sequencing is helping scientists find accurate and actionable genome-wide insights into their research. Dr. Liu will share how targeted haplotype-resolved assembly provides insights into allele-specific regulation of the MHC region at this webinar.

It is well known that the major histocompatibility complex (MHC) region of the human genome is 4.5 Mb long and is one of the most complex and difficult to study regions of the human genome. Dr. Yun Liu and his team from the School of Basic Medicine, Fudan University, used a combination of CRISPR-based targeted enrichment strategy + 10X genomics and PacBio HiFi sequencing to detect enrichment with only 20ng of DNA input. The captured regions were subjected to 30-fold HiFi sequencing.

Key takeaways of this webinar include:

  1. The targeted approach greatly reduced the required input and resources for achieving haplotype-resolved assembly.
  2. Compared to the results from haplotype-resolved genome assembly, the targeted approach achieved comparable completeness and accuracy, enabling comprehensive detection of genetic variants of the targeted region, such as the MHC.
  3. Using the targeted assembled personal haplotypes as the reference not only improves the quantification accuracy for sequencing data but also enables allele-specific functional genomics analyses

Professor Yun LIU, Fudan University

Prof. Liu obtained his B.S. in Biology from Fudan University in 2003. He received the MHS degree in Biostatistics and the Ph.D. degree in Molecular Biology and Genetics, both from Johns Hopkins University in 2009. He then did his postdoctoral training in Johns Hopkins University Center for Epigenetics. In 2014, Prof. Liu joined Fudan University to set up his own lab. He is currently a professor in the Department of Biochemistry & Molecular Biology in Fudan University School of Basic Medical Sciences. Prof. Liu’s laboratory is interested in the roles of epigenetic regulations in human common diseases and cancers. His works were published on a lot of high-profile scientific journals, such as Nature Biotechnology, Nature Genetics, American Journal of Human Genetics and Nucleic Acids Research, etc. His papers were well-recognized in the scientific community and were cited more than 3000 times in the past 5 years.
